Objective: Guided by the theory of decaying and generating muscle,to observe the effect of Xiaoyanshengji ointment on promoting wound healing in rats, and to investigate the mechanism of Xiaoyanshengji ointment in promoting wound healing by detecting the expression of IL-6 and IFN-γin the process of wound healing.Method:Sixty male SD rats were randomly divided into blank control group, Xiaoyan Shengji ointment group, positive drug erythromycin ointment group and saline group, there are 12 subgroups with a total of 60 birds.The wound healing was observed and the granulation tissue was obtained after the animals were killed on the 3rd, 7th and 14th day.The histomorphological changes of the wounds were observed by HE staining and the mRNA and protein expressions of IL-6 and IFN-γwere detected by RP-PCR and immunohistochemistry.Results:
The wound healing area was (70.20±16.72) mm2 in Xiaoyanshengji ointment group, which was significantly smaller than that in normal saline group (112.0±4.24) mm2(F= 26.862, P = 0.001);Under light microscope, a large number of fibrocytes were found in the visual field on the 14th day after treatment with Xiaoyan Shengji ointment;The expression of IL-6 in erythromycin group and Xiaoyanshengji ointment group were (9.72±0.47) pg/mL and (11.83±0.83) pg/mL respectively on the 14th day after treatment, the difference was significant (F= 155.791; P = 0.000) The expression of IFN-γin erythromycin group and Xiaoyanshengji ointment group were (352.94±16.09) pg/mL and (377.61±15.43) pg/mL, respectively, without statistical significance (F= 52.093; P= 0.000);The results of RT-PCR showed that there were significant differences in IL-6 mRNA and IFN-γmRNA among the groups on the 14th day (F = 121.078; P = 0.000; F = 369.189; P = 0.000 ) The expression of IL-6 mrna in erythromycin group and Xiaoyanshengji ointment group were (2.69±0.20) and (4.04±0.62) , respectively, with significant difference (P = 0.030) The expression of IFN-γin erythromycin group and Xiaoyanshengji ointment group were (3.16±0.15) and (3.25±0.14) , respectively, with no statistical significance (P = 0.431);The expression of IL-6 and IFN-γin the normal saline group, erythromycin group and Xiaoyanshengji ointment group was higher than that in the blank control group, but the expression of IL-6 and IFN-γin the erythromycin group and Xiaoyanshengji ointment group was higher than that in the normal control group, the expression levels of IL-6 and IFN-γgradually decreased with time.Conclusion: The mechanism of Yanshengji ointment in promoting wound healing may be related to the down-regulation of the expression of IL-6 and IFN-γin the wound tissue. |
